While I agree that the problem seems to be #20372 this workaround worked for me.. I'm on running OsX 10.4.11 on PPC. I have kept the default for universal builds. Note that both glib2 and gtk2 were built and working last month, something changed somewhere.

GTK+ 3 will overwrite gtk-update-icon-cache from GTK+-2.24.32 if it is installed. There is nothing wrong about that assuming that both programs provide same functionality. If you wish to keep one from GTK+ 2 you can add --enable-gtk2-dependency to the configure command.
